Testosterone taken by mouth is extensively broken down by the liver before it can reach circulation, which is why most oral formulations have struggled to deliver consistent, predictable exposure. That first-pass metabolism is the central obstacle STC-101 is designed to work with, not around.
STC-101 is an oral prodrug designed to activate in the liver and release testosterone, using the body's own metabolic machinery rather than working against it. The goal is a practical, once-daily therapy with more predictable exposure, built through a stepwise, data-driven process across computational, in vitro, and in vivo studies.
Preclinical proof-of-concept.
Preclinical work to date spans computational modeling, liver-cell studies, and animal pharmacokinetics, each testing a different part of how STC-101 is designed to work.
